How it works

Azathioprine inhibits purine synthesis. Purines are needed to produce DNA and RNA. By inhibiting purine synthesis, less DNA and RNA are produced for the synthesis of white blood cells, thus causing immunosuppression.

Side Effects

General tiredness, dizziness, feeling sick (nausea), being sick (vomiting), diarrhoea or abdominal pain , swelling of the eyelids, face or lips redness of the skin, skin nodules or a skin rash (including blisters, itching or peeling skin) , pain in the muscles or joints , sudden wheeziness, coughing or difficulty breathing .

Drug Interactions

Ciclosporin, methotrexate, tacrolimus , doxorubicin , allopurinol, oxipurinol or thiopurinol , cimetidine , furosemide , penicillamine , anticoagulants such as warfarin or acenocoumarol , captopril , indomethacin , co-trimoxazole , infliximab, mesalazine, olsalazine, sulfasalazine or balsalazide , tubocurarine or succinylcholine .

Indication

It is indicated as an adjunct for the prevention of rejection in renal homotransplantation. It is also indicated for the management of active rheumatoid arthritis to reduce signs and symptoms.

When not to Use

It should not be given to patients who have shown hypersensitivity to the drug. It should not be used for treating rheumatoid arthritis in pregnant women. Patients with rheumatoid arthritis previously treated with alkylating agents (cyclophosphamide, chlorambucil, melphalan, or others) may have a prohibitive risk of malignancy if treated with this medicine.

Amorin Tablets Precautions

Precaution

A gastrointestinal hypersensitivity reaction characterized by severe nausea and vomiting has been reported. These symptoms may also be accompanied by diarrhea, rash, fever, malaise, myalgias, elevations in liver enzymes, and occasionally, hypotension. Symptoms of gastrointestinal toxicity most often develop within the first several weeks of therapy with azathioprine and are reversible upon discontinuation of the drug. The reaction can recur within hours after re-challenge with a single dose of azathioprine.

Amorin Tablets Warnings

Warning 1

Patients receiving immunosuppressants, including azathioprine, are at increased risk of developing lymphoma and other malignancies, particularly of the skin. Physicians should inform patients of the risk of malignancy with azathioprine. As usual for patients with increased risk for skin cancer, exposure to sunlight and ultraviolet light should be limited by wearing protective clothing and using a sunscreen with a high protection factor.

Warning 2

Serious infections are a constant hazard for patients receiving chronic immunosuppression, especially for homograft recipients. Fungal, viral, bacterial, and protozoal infections may be fatal and should be treated vigorously. Reduction of azathioprine dosage and/or use of other drugs should be considered.

Warning 3

If you have an inherited mutation in the NUDT15-gene (a gene which is involved in the break-down of Azathioprine in the body), you have a higher risk of infections and hair loss and your doctor may in this case give you a lower dose.
